What is the procedure to test the gain of antenna in nRF52840 DK or can you please tell the value of gain of antenna in nRF52840 DK?

The antenna gain on the DK will have a gain of about 0 dBi.

There are various articles on the Web on antenna testing. Here is a good one. www.testandmeasurementtips.com/.../
